Synopsis

This course is part of a series of three post-congress courses from the World Muscle Society (WMS), designed to translate the most relevant scientific advances in the treatment of neuromuscular diseases into clinical practice. This third course focuses on advances in emerging therapies and innovative treatment strategies, with particular emphasis on current challenges in therapeutic delivery, immune response modulation, and the development of new technological platforms. Based on the key messages presented at the congress, it addresses core concepts that are redefining therapeutic approaches in these diseases. The programme explores the current limitations of gene therapy, including vector efficiency, toxicity related to viral load, and immune responses, as well as new strategies aimed at improving tissue selectivity and reducing adverse effects. It also reviews innovative approaches such as the use of nanoparticles, vector engineering, combination therapy with immunomodulators, and the development of advanced human models for translational research. In addition, new pathophysiological concepts and relevant clinical examples are incorporated, illustrating how molecular knowledge is directly impacting the design of new therapies. This course highlights that, beyond diagnosis, the future of managing these diseases lies in the ability to personalise and optimise therapeutic strategies.
