Synopsis

HER2-mutant non-small cell lung cancer (NSCLC) represents a distinct molecular subgroup for which the treatment landscape is rapidly evolving. This case-based educational activity uses two patient journeys to explore key considerations in the management of HER2-mutant advanced NSCLC, from molecular identification and treatment selection to the management of disease progression and central nervous system involvement. Through two complementary clinical cases, the faculty review current evidence for HER2-directed therapies and consider how efficacy, intracranial activity, safety and patient and disease characteristics may inform treatment decisions. Particular attention is given to the management of symptomatic and asymptomatic brain metastases, the integration of local and systemic treatment approaches, and appropriate monitoring for treatment-related adverse events. The cases also provide a framework for discussing emerging questions around treatment sequencing and mechanisms of resistance to HER2-directed therapies, highlighting the importance of evidence-based, individualised and multidisciplinary decision-making as the therapeutic landscape continues to evolve. The funding for this educational program is supported by a grant from Daiichi Sankyo Europe (DSE) and AstraZeneca (AZ). DSE and AZ had no involvement in the design, agenda, selection of speakers and participants

Objectives

After completing this activity, participants should be better prepared to:

  • Evaluate current evidence for HER2-directed treatment strategies in patients with HER2-mutant advanced NSCLC.
  • Apply evidence-based principles to the management of brain metastases in HER2-mutant NSCLC, considering neurological symptoms, local treatment options and the intracranial activity of systemic therapies.
  • Recognise key considerations influencing treatment selection and sequencing, including efficacy, safety, disease progression and emerging mechanisms of resistance.
